Mandalay Bay's location pretty much forces you to use some kind of transit to get anywhere else unless you want to go to maybe Luxor or Excalibur. MGM Grand is so damn huge, it can take you 35 hours to walk from your room to the Strip (I exaggerate only slightly). I also like NYNY for the somewhat more quirky atmosphere--it's a bit more thematic.
I must confess that my primary objective, if I was staying on the Strip, would be to find the best way to get the hell off of it and get to a non-gouge casino. So NYNY or MGM wins there, as I could take the bus to the Orleans easily enough from that corner--or the Strip bus downtown.
If "where does the gambling suck least" is a factor, it's pretty much a three-way tie there, sad to say. Same with value and quality of food (awful for what you get). Squeeze your freebies, maybe give them enough play to get more (it won't be much more), and concentrate future efforts away from the Strip megatoilets.
