Suite vs Regular Room

I have stayed in a standard room everywhere on your list except Cosmo. I have found that a great room at a good hotel does not always equate to a great experiance. I would go standard room at Wynn or Cosmo.
It depends on the hotel. For instance, I've stayed in a on bedroom suite at Luxor. While very large, it was short on amenities and not all that nice. A regular room at Palazzo or Wynn is nicer than the Luxor Suite.

OTOH the regular rooms at the Hotel at Mandalay Bay are all one bedroom suites and quite nice. I like them lots and would take a basic room at the Hotel over any of Mandalay Bay's lower tier suite rooms.

So the answer is that it depends.
Regular room at a swanky hotel. Hands down. I've found "suites" in some hotels to be a large regular room.
I've found the rooms at Wynn and the suites at Encore are about the same. Only a partition between the rooms make one a suite and another a room.

To me, a true suite should consist of 2 full rooms. I've stayed in suites at Silverton and RR which were actually quite nice. I consider the pretty rooms at V/P semi-suites in that there is just a step down to the living room but it's still one room. I've never been in a higher tier suite so I can't say if I'd prefer those vs. a newer, regular room at the nicer hotels. Would be pretty cool to stay in a Villa at CP, do some bowling, add some blow up toys to the tub, perhaps keep a baby in the closet and a tiger in the bathroom...
I agree with wanabe on suite defenition. Personally, as I usually travel solo, a regular room with a decent chair or a mini suite with a seating area is all I need. On a trip a while back. I was upgraded to an actual suite after a hotel screwup. It was fine, but other than my locals friends stopping by for a drink to see it, it was wasted. I would have preferred a comp on a standard room. When I'm solo, a nice room like GVR or RR would be all I needed.

I've never stayed in a Vegas Suite. Although if I ever had one, it has to contain a hot tub.

I don't know if I know of any suites with a hot tub

One of these days, I'm going to splurge on one of these Palms Fantasy Suites....

Luxor has those corner rooms with a tub. I'm not sure where you draw the line between a big Jacuzzi tub and a hot tub. Maybe a hot tub has to sit at least four people?
